Western blot
Brain tissue from WT and COMT−/− mice. Tissues were lysed in ice-cold RIPA buffer (Teknova) with 10 mm PMSF (Fluka) and Complete Protease Inhibitor Cocktail (Roche).
5% nonfat dry milk in buffer containing 1 M Tris-buffered saline and 0.05% Tween for 30 min.
1:1,000 dilution overnight at 4°C.
1:5,000 dilution in goat anti-mouse peroxidase-conjugated (Jackson ImmunoResearch) for 1 h at room temperature.
Enhanced chemiluminescence (PerkinElmer) and exposed to film (Denville Scientific).
