Brain from Chn1WT/WT, Chn1WT/KO and Chn1KO/KO mice. Postnatal day 0-2 whole brain was homogenized on ice in RIPA buffer (Thermo Scientific), 1mM EDTA (Gibco), and Halt protease and phosphatase inhibitor cocktail (Thermo Scientific).

Blocking agent

5% milk in PBST.

Primary incubation

1:1,000 dilution overnight at 4 C.

Secondary incubation

1:5,000 dilution peroxidase AffiniPure donkey anti-rabbit IgG (Jackson ImmunoResearch).

Detection

Amersham ECL Prime Western Blotting Detection Reagent (GE Healthcare) and imaged with a FujiFilm LAS-4000 with CCD camera (GE Healthcare) and MultiGauge software (GE Healthcare).
