Sample

WB: cultured fetal-derived NSCs from Chd7null (NestinCre;Chd7-/-) and control (Chd7+/+) mouse cells lysed in 8 M urea, 1% CHAPS, 50 mM TRIS pH7.9.

IHC: Coronal mouse brain sections (20-µm-thick) from GLAST::CreERT2;Chd7f/f mouse.

Blocking agent

WB: 0.5% nonfat milk powder in TBS with 0.5% Tween 20 (TBST).

IHC: 10% heat-inactivated goat serum (GS) for 1 hour.

Primary incubation

WB: 1/2,000 in 5% BSA, TBST overnight at 4°C.

IHC: 1/80 in 5% GS in 0.1% Triton X (Sigma) in PBS overnight at 4°C.

Secondary incubation

WB: horseradish peroxidase (HRP)-conjugated secondary antibody (Millipore, U.K.) for 1 hour at room temperature.

IHC: Alexa-Fluor-labeled secondary antibodies diluted at 1/200 (Life Technologies) for 1 hour.

Detection

WB: Clarity Western ECL reagent (Bio-rad, U.K.) and a Bio-Rad gel doc system.
