CD44 antibody | knockout validation | BD Biosciences 553131

This is a knockout-validated antibody summary, based on the publication "Epidermal-specific deletion of CD44 reveals a function in keratinocytes in response to mechanical stress", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

CD44 antibody | knockout validation | BD Biosciences 553131 figure 1
Figure 1. A pan CD44 antibody (IM7; red, green) was used for indirect immune fluorescence staining of skin sections or keratinocytes (lower panel) harvested from Cd44Δker mice or control animals. Nuclei were counterstained with DAPI (blue). White arrows indicate areas where CD44 is removed (epidermis, hair follicles). From [1].
Antibody information

Rat monoclonal IgG2b

Company: BD Biosciences

Antibody: CD44

Catalog number: 553131

Summary: Rat monoclonal IgG2b against dexamethasone-induced SJL mouse spontaneous myeloid leukemia M1 cells. Reacts with mouse by flow cytometry,

immunoprecipitation, ELISA, radioimmunoassay, immunohistochemistry (paraffin and frozen), cytotoxicity, blocking-activation assays.

Validation Method

Immunohistochemistry | Immunocytochemistry

Sample

Skin sections (frozen or paraffin) and keratinocytes from control and Cd44Δker mice.

Blocking agent

5% FCS in PBS for 1 h at room temperature.

Primary incubation

2 μg/ml in blocking solution overnight.

Secondary incubation

1:500 dilution Alexa Fluor labeled secondary antibodies in blocking solution for 1 h at room temperature.

Notes

Antibodies pan CD44 antibody (KM201) or a CD44v6 specific antibody (α9A4) are KO validated by IHC and WB (Fig 1b,c), but Authors do not specify the manufacturer in the article. The author indicated that the KM201 Ab was from Abcam (ab25340). The 9A4 is prepared by authors. The hybridoma cells 9A4 can be found in ATCC.
