Summary: Rat monoclonal IgG2a raised against IL-4 treated murine thioglycollate-elicited peritoneal macrophages. It detects mouse CD36 protein recommended for flow cytometry and immunofluorescence.

Validation Method

Immunohistochemistry

Sample

Head sections (olfactory epithelium) of CD36−/− mice. The head was fixed for 2 h at 4°C in fixative solution, cryoprotected in 30% sucrose, and embedded in Leica tissue freezing medium (Leica Microsystems). Sections of 12 μm thickness were cut on a Leica CM 1900 UV cryostat (Leica Microsystems), adhered to Super Frost® Plus cryoslides (Thermo Scientific) and stored at −25°C.

Blocking agent

1x PBS+/+ (PBS + 0.9 mM CaCl2, 0.48 mM MgCl2), 0.1% Triton X-100, 1% gelatin (Sigma, G7765) for at least 1 h

Primary incubation

1:200, overnight at 4°C

Secondary incubation

1:500 secondary antibody for 1 h

Detection

confocal microscopy with TCS SPE and TCS SP5 systems (Leica microsystems)
