CD274 (PD-L1, B7-H1) antibody | knockout validation | eBiosciences 14-5983-82

This is a knockout-validated antibody summary, based on the publication "CMTM6 maintains the expression of PD-L1 and regulates anti-tumour immunity", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Validation Method

Immunocytochemistry

Sample

MDA-MB-231 cells

Blocking agent

5% BSA, 0.3M glycine in PBST for 1 hour.

Primary incubation

At 4° C overnight.

Secondary incubation

Fluorescent conjugated secondary antibody for 1 h at RT.

Detection

Nikon C2 confocal microscope.

Figure

PD-L1 and CMTM6 co-localise at the plasma membrane. PD-L1 knockout, CMTM6 knockout and parental MDA-MB-231 Cas9 cells were fixed, immunostained for PD-L1 and CMTM6 and analysed by confocal microscopy. Scale bar: 10μm. Please see Figure 2c in the article [1].
