CD163 antibody | knockout validation | Bio-Rad MCA2311GA

This is a knockout-validated antibody summary, based on the publication "CD163 knockout pigs are fully resistant to highly pathogenic porcine reproductive and respiratory syndrome virus", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG

Company: Bio-Rad

Antibody: CD163

Catalog number: MCA2311GA

Summary: Mouse monoclonal IgG against CD163 from porcine alveolar macrophages. Reacts with porcine by western blot, Immunoprecipitation, immunohistochemistry (frozen), immunofluorescence and flow cytometry.

Validation Method

Western blot

Sample

Lung tissues from CD163 KO pigs.

Blocking agent

5% non-fat dry milk.

Secondary incubation

HRP-conjugated anti-mouse IgG (ab6789, Abcam).

Detection

Pierce™ ECL Western Blotting Substrate (Thermo Fisher Scientific). Chemiluminescent signals were captured by a cooled charged-coupled device (CCD) camera.

Figure

Western blot showing the CD163 protein expression is undetectable in the lungs of KO pigs. Two antibodies that recognized non-reducing (upper panel) and reducing CD163 (middle panel) proteins, respectively, were used for detecting CD163, and β-actin was stained as loading control (lower panel). CD163 protein possessed a molecular mass of approximately 130 kDa. A higher-molecular-mass band of unknown identity was also detected in the lungs of pigs. Please see Figure 1c in the article [1].

Clone note

The same clone (2A10/11) is sold as Bio-Rad MCA2311F, MCA2311PE, MCA2311GA; Invitrogen MA5-16476, MA5-28292, MA5-28293.
