Immunocytochemistry

Sample

WT and different domain deletion BubR1 MEFs. Cells were fixed in 3% paraformaldehyde.

Blocking agent

PBS–2% nonfat milk (Kroger Co., Cincinnati, Ohio).

Primary incubation

1:250 dilution overnight in blocking buffer.

Secondary incubation

Texas red-conjugated goat anti-mouse antibodies at 4 μg per ml in blocking buffer.

Detection

Laser-scanning microscope (LSM 880; Carl Zeiss) with an inverted microscope (Axiovert 100 M; Carl Zeiss).
