This is a knockout-validated antibody summary, based on the publication "The Tec Kinase-Regulated Phosphoproteome Reveals a Mechanism for the Regulation of Inhibitory Signals in Murine Macrophages", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG2a

Company: BD Biosciences

Antibody: Btk

Catalog number: 558528

Summary: Mouse monoclonal IgG2a against a recombinant protein corresponding to human N-Terminal Btk (aa. 2-172). Reacts with human by western blot and immunofluorescence.

Validation Method

Western blot

Sample

Bone marrow-derived macrophages from wild-type, Btk-/- or Btk-/-Tec-/- mice. Whole cell protein extracts were prepared by cell lysis with buffer containing 50mM Tris pH 7.4, 150mM sodium chloride, 1mM EDTA, 1% Triton X-100, 1mM sodium orthovanadate, 0.25% sodium deoxycholate and protease inhibitors.

Detection

Odyssey infrared imaging system software (Licor Biosciences, Lincoln, NE) (Li-Cor).

Figure

Western blot of the indicated proteins in bone marrow-derived macrophages from wild-type, Btk-/- or Btk-/-Tec-/- mice were left unstimulated or stimulated with LPS (1 ng/ml) for the indicated times (min). Please see Figure S2b in the article [1].
