Validation Method

Western blot | Immunocytochemistry

Sample

Control and (Bax/Bak)-/- HCT-116 cells.

Blocking agent

5% non-fat dry milk in Tris-buffered saline with Tween 20 (TBST) (15 mm Tris-HCl, pH 7.5, 200 mm NaCl, and 0.1% Tween 20) at room temperature for 1 h.

Primary incubation

At room temperature for 2 h or overnight at 4˚C.

Secondary incubation

Peroxidase-conjugated secondary antibodies (Jackson Laboratories) for 1 h.

Detection

Enhanced chemiluminescence detection reagent (Millipore, Ireland).
