Sample

Mouse tissues from control and liver specific BVRA-knockout mice. WB: Tissues were lysed in CelLytic Buffer (Sigma C3228) plus 10% protease inhibitor cocktail (Sigma P2714-1BTL) and Halt phosphatase inhibitor cocktail (Fisher PI78420). IHC: Liver sections were fixed in paraformaldehyde.

Blocking agent

WB: 3% BSA in TBS (10 mM Tris-HCl (pH 7.4) and 150 mM NaCl) at room temperature for 2 h.

IHC: 5% normal donkey serum (NDS) for 1 h at 4°C.

Primary incubation

WB: overnight at 4°C.

IHC: 1:100 dilution in 5% NDS overnight at 4°C.

Secondary incubation

WB: 1:10,000 dilution infrared anti-rabbit labeled with IRDye 800 (LI-COR Biosciences) in TBS for 2 h at 4°C.

IHC: 1:1, 000 dilution fluorescent-labeled donkey anti-rabbit Alexa488 (Invitrogen, A-21206).

Detection

WB: infrared scanning with Odyssey system (LI-COR Biosciences).

IHC: Leica TCS SP5 Laser Scanning Confocal Microscope (Leica Microsystems, Buffalo Grove, IL).
