This is a knockout-validated antibody summary, based on the publication "Role of PINCH and its partner tumor suppressor Rsu-1 in regulating liver size and tumorigenesis", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Company: BD

Antibody: PINCH 1

Catalog number: 612710

Summary

Mouse IgG2a monoclonal antibody raised against human PINCH peptide (aa. 120-220). It reacts to human, mouse, rat, and dog. It has been successfully used for western blot.

BD mouse monoclonal PINCH 1 antibody 612710 was validated in knockout mice figure 1
Figure 1. Western blot of PINCH 1 on hepatocytes isolated from PINCH1 and PINCH 2 double knockout mice and control mice. Actin is used as a load control. from Figure 2 [1].
Validation Method

WB, western blot

Sample

Hepatocytes from PINCH 1 and PINCH 2 double knockout (PINCH DKO) mice. Total protein was isolated from the mouse hepatocytes or whole livers of PINCH DKO and control mice using 1% sodium dodecyl sulfate (SDS) in RIPA buffer (20 mM Tris/Cl pH 7.5, 150 mM NaCl, 0.5% NP-40, 1% TX-100, 0.25% Sodium Deoxycholate (DOC), 0.6-2 µg/ml aprotinin, 10µM Leupeptin, 1µM Pepstatin).

Primary incubation

Tris-buffered saline Tween 20 containing 5% nonfat milk

Secondary incubation

Tris-buffered saline Tween 20 containing 5% nonfat milk

Detection

SuperSignal West Pico chemiluminescence substrate (Pierce, Rockford, IL) and exposed to a X-ray film (Lab Product, Sales, Rochester, NY).
