Sample

Bone marrow stromal cells and bone sections from wild-type and Anxa2 knocked-out mice. IC: Cells were fixed with 10% neutral buffered formalin (cat. HT501320, Sigma) and permeabilized with PBT (1:500 dilution of Triton-X100 in PBS). IHC: long bones were embedded in paraffin for sectioning.

Blocking agent

IHC: Image-iT FX signal enhancer (cat. I36933, Life Technology, Madison, WI) for 30 min.

Primary incubation

1:25 dilution for 2 hours at room temperature.

Secondary incubation

Primary antibodies were combined with reagents of Zenon Alexa Fluor 488 (green) or 555 (red) labeling kit (Life Technology).

Detection

Olympus FV-500 confocal microscope.
