Ah Receptor antibody | knockout validation | Santa Cruz sc-398877

This is a knockout-validated antibody summary, based on the publication "Neurons expressing the aryl hydrocarbon receptor in the locus coeruleus and island of Calleja major are novel targets of dioxin in the mouse brain", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Figure 1. Images showing AhR and GAPDH proteins detected by western blotting in the brain, liver, and lung of Ahr+/+ and Ahr−/− mice at P5 (n=1 in each genotype). No AhR protein was detected in these tissues of Ahr−/− mice, demonstrating the specificity of the antibody. From [1].
Antibody information

Mouse monoclonal IgG1 κ

Company: Santa Cruz

Antibody: Ah Receptor

Catalog number: sc-398877

Summary: Mouse monoclonal antibody specific for an epitope mapping between amino acids 767-800 at the C-terminus of mouse Ah Receptor. Recommended for detection of Ah Receptor of mouse and rat origin by western blot, immunoprecipitation, immunofluorescence, immunohistochemistry (paraffin) and ELISA.

Validation Method

Figure 2. Representative images showing the locus coeruleus (LC) of Ahr+/+ and Ahr−/− mice at P14 (n=3 mice in each genotype), demonstrating antibody specificity. Arrowheads mark TH-negative cells with nonspecific anti-AhR antibody staining. Scale bar=100 μm. From [1].
Sample

Control and Ahr−/− mice tissues.

Blocking agent

IHC: PBST containing 5% bovine serum albumin (A3059; Sigma-Aldrich).

Primary incubation

WB: 1:1,000 dilution in Tris-bufered saline, pH 7.4, containing 0.1% Tween-20 (TBST), overnight at 4 °C.

IHC: 1:500 dilution in blocking solution overnight at 4 °C.

Secondary incubation

WB: 1:5,000 dilution anti-mouse IgG-horseradish peroxidase (HRP)-conjugated antibody (7076S, Cell Signaling Technology, Beverly, MA, USA) in TBST for 1 h at room temperature.

IHC: 1:1,000 dilution anti mouse IgG AlexaFluor 488 (Life Technologies, Gaithersburg, MD, USA) in PBST.

Detection

WB: Chemi-Lumi One (Nacalai Tesque, Kyoto, Japan) was used to visualize the protein bands, which were detected on Hyperfilm ECL (GE Healthcare Ltd., Tokyo, Japan) and developed and fixed with GBX developer and fixer (Kodak, Rochester, NY, USA).

IHC: inverted Leica DMi8 microscope, equipped with the Leica TCS SP8 confocal module (Leica Microsystems).
