Abl antibody | knockout validation | BD Pharmingen 8E9

This is a knockout-validated antibody summary, based on the publications "A crucial role in cell spreading for the interaction of Abl PxxP motifs with Crk and Nck adaptors" [1] and "Activation of tyrosine kinase c-Abl contributes to α-synuclein-induced neurodegeneration" [2]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1

Company: BD Pharmingen

Antibody: Abl

Catalog number: 8E9

Summary: Mouse monoclonal IgG1 against recombinant mouse Abl Gag fusion protein. Reacts with human and mouse protein. Suitable for western blot, immunofluorescence and immunoprecipitation.

in article "A crucial role in cell spreading for the interaction of Abl PxxP motifs with Crk and Nck adaptors"
Validation Method

Immunocytochemistry

Sample

Mouse embryonic fibroblasts lacking Abl and Arg (Abl DKO cells) stably expressing various forms of Abl. Cells were fixed on fibronectin-coated glass coverslips using 37% (w/v) formaldehyde.

Blocking agent

1% (w/v) BSA in PBS for 1 hour at room temperature.

Secondary incubation

Goat anti-mouse secondary antibody conjugated with Alexa-fluor 488 (Invitrogen).

Detection

Photomicrographs were obtained on a Zeiss 510 laser-scanning confocal microscope.

Figure

Please see Figure 1b in the article [1].

Western blot validation in article "Activation of tyrosine kinase c-Abl contributes to α-synuclein-induced neurodegeneration"

This antibody is validated for Western blot in the detergent-soluble fraction of brain stem from 9-month-old c-AblWT hA53Tα-syn transgenic mice versus c-AblKO hA53Tα-syn transgenic mice and age-matched littermate controls. See figure 5D in [2].
