ATM antibody | knockout validation | Abcam ab78

This is a knockout-validated antibody summary, based on the publication "Loss of ATM accelerates pancreatic cancer formation and epithelial-mesenchymal transition" [2] (discussed below) and "CRISPaint allows modular base-specific gene tagging using a ligase-4-dependent mechanism" [1] (see Figure 1 or figure 6b in the article). Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1

Company: Abcam

Antibody: ATM

Catalog number: ab78

Summary: Mouse monoclonal IgG1 against a fusion protein corresponding to amino acids 2577-3056 of human protein. Reacts with mouse, rat, human, and monkey. Suitable for western blot, immunoprecipitation, immunofluorescence, immunohistochemistry (paraffin) and flow cytometry.

Validation Method

Western blot

Sample

Mice pancreas from mice harbouring a floxed Atm (‘A'), p48Cre/+, and KrasG12D/+ (‘K'). Pancreas were homogenized in extraction buffer (4% SDS, 100 mM Tris-HCl) containing protease inhibitors and 1 mM phenylmethylsulfonyl fluoride (PMSF),

Primary incubation

1:1000 dilution overnight at 4° C.

Detection

Immunoreactive bands were visualized using chemiluminescence (Thermo scientific, Waltham, MA, USA).
