This is a knockout-validated antibody summary, based on the publication "Adipose triglyceride lipase in human skeletal muscle is upregulated by exercise training", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Rabbit polyclonal

Company: Cell Signaling

Antibody: ATGL

Catalog number: 2138

Summary: Rabbit polyclonal against a synthetic peptide corresponding to a sequence around Pro186 of human ATGL. Reacts with mouse and is predicted to react based on 100% sequence homology with rat. Suitable for western blotting, immunoprecipitation, immunohistochemistry (paraffin), and immunofluorescence/immunocytochemistry.

Validation Method

Western blot

Sample

Skeletal muscle lysates from ATGL knockout and wild-type mice lysates. Muscle tissue was homogenized, while kept at ∼0°C in an ice slurry in a buffer (1:80 wt:vol) containing 50 mM HEPES pH 7.5, 150 mM NaCl, 20 mM Na pyrophosphate, 20 mM β-glycerophosphate, 10 mM Na fluoride, 2 mM Na orthovanadate, 2 mM EDTA, 1 mM EGTA, 1% Nonidet P-40, 10% glycerol, 2 mM PMSF, 10 μg/ml leupeptin, 10 μg/ml aprotinin, and 3 mM benzamidine using a polytron homogenizer (PT 1200; Kinematica, Luzern, Switzerland).

Blocking agent

2% skimmed milk or 5% BSA in Tris-buffered saline containing 0.1% Tween for 2 h at room temperature.

Primary incubation

Overnight at 4°C.

Secondary incubation

Horseradish peroxidase-conjugated anti-rabbit (cat. no. P0448; DAKO; Glostrup, Denmark).

Detection

Enhanced chemiluminescence (Immobilion Western; Millipore) and Kodak Image station 2000MM (Kodak, Glostrup, Denmark).

Disclaimer

If the antibody described in this summary is a polyclonal antibody, since polyclonal antibodies are of limited quantity, please inquire the supplier whether any current polyclonal antibody with the same catalog number is exactly the same as the one described in this summary. Sometimes, different bleeds or different animals are used, usually with a different lot number. In such cases, the result in this summary may not apply to the new antibody with the same catalog number.
