Sample

Liver from control and ASL-KO mice. WB: Tissue from 3 mice per group was homogenized in RIPA buffer (Sigma, catalog no. R0278-50 mL) containing proteases inhibitors (Roche, Basel, Switzerland, catalog no. 04693159001). IHC: Paraffin sections were treated with Antigen Retrieval Solution (Dako, Copenhagen, Denmark, catalog no. S236984) for 30 minutes.

Blocking agent

WB: 3% bovine serum albumin.

IHC: 3% hydrogen peroxide for 5 minutes.

Primary incubation

WB: 1:500 dilution.

IHC: 1:100 dilution at 4°C overnight.

Secondary incubation

WB: 1:20,000 dilution donkey anti-mouse immunoglobulin G/horseradish peroxidase (Jackson ImmunoResearch, West Grove, PA, catalog no.715-035-150).

IHC: M.O.M Vectastain and DAB detection kit (Vector Laboratories, Burlingame, CA, catalog no. PK-2200, and SK-4100, respectively) according to the manufacturer’s recommendations.

Detection

WB: Amersham ECL Western Blotting Detection Reagent (General Electric Healthcare Life Sciences, Marlborough, MA, catalog no. RPN2106).
