Summary: Rabbit monoclonal IgG against a synthetic peptide within human androgen receptor aa 900 to the C-terminus. Reacts with human, mouse and rat. Suitable for western blot. Unsuitable for flow cytometry, immunocytochemistry/immunofluorescence or immunohistochemistry (paraffin).

Immunohistochemistry

Brain tissue from wildtype and AR-knockout mice. Brains were post-fixed for 2 hours in 4% paraformaldehyde, at 4°C, incubated in a 20% sucrose solution and microtome sectioned in the coronal plane at 35µm.

10% NGS in PBS-GT (0.1% gelatin, 0.3% Triton X-100, in PBS, pH 7.4) for 1 hour and biotin block (Avidin/Biotin Blocking Kit, Vector, cat#SP-2001) for 10 minutes.

1:5,000 dilution in 1% NGS in PBS-GT for 36 hours at 4°C.

1:1,000 dilution biotinylated goat anti-rabbit secondary antisera (Jackson Immunoresearch, lot # 88762, code # 711-065-152) in 1% NGS in PBS-GT for 1 hour .

Avidin-Biotin complex solution made 30 minutes before use (1 drop of each solution A and B per 10ml of PBS-GT; ABC Elite Kit, Vector Laboratories, catalogue # PK6100) for 1 hour and NiCl-enhanced diaminobenzidine (DAB, Sigma, St. Louis, MO) in a 0.05 M Tris Buffer, pH 7.2.

The manufacturer (Abcam) does not recommend this Ab for IHC, but it was used successfully.
