Validation Method

Western blot | Immunohistochemistry

Sample

Brains from WT and ApoE-knockout mice. WB: tissues were lysed in RIPA Lysis Buffer with a protease inhibitor cocktail (05892791001; Roche, Basel, Switzerland) and homogenized at 4°C until lysis was complete. IHC: mice were perfused with 4% paraformaldehyde (PFA), the whole brains were harvested and coronary frozen sections were made.

Blocking agent

WB: 5% bovine serum albumin at room temperature for 1 h.

Primary incubation

WB: At 4°C overnight.

Secondary incubation

WB: for 1 h.

IHC: DyLight 594 AffiniPure Goat Anti-Mouse IgG (A23410, Abbkine).

Detection

WB: ECL Western Blotting kit (32209; Thermo Scientific, Waltham, MA) and chemiluminescence imaging system (ChemiDoc XRS+; Bio-Rad, Hercules, CA).
