Hindlimb muscles from genotyped Ano5+/+, Ano5+/− and Ano5−/− mice. Tissues were homogenized in 50 mm Bis–Tris, 6N HCl, 50 mM NaCl, 10% w/v glycerol, 0.001% ponceau S, pH 7.2 supplemented with protease inhibitor cocktail III (Calbiochem).
5% non-fat dry milk for 1 h.
1:100 dilution in PBST + 1% milk overnight at 4°C.
HRP conjugated goat anti-mouse (Biorad) for 1 h at room temperature.
ECL and radiographic film.
