ANO5 antibody | knockout validation | NeuroMab 75-410

This is a knockout-validated antibody summary, based on the publication "Defective membrane fusion and repair in Anoctamin5-deficient muscular dystrophy", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Antibody information

Mouse monoclonal IgG1

Company: NeuroMab

Antibody: ANO5

Catalog number: 75-410

Summary: Mouse monoclonal IgG1 against a fusion protein corresponding to amino acids 1-299 (cytoplasmic N-terminus) of human ANO5. Reacts with human by immunoblot and immunocytochemistry.

Validation Method

Western blot

Sample

Hindlimb muscles from genotyped Ano5+/+, Ano5+/− and Ano5−/− mice. Tissues were homogenized in 50 mm Bis–Tris, 6N HCl, 50 mM NaCl, 10% w/v glycerol, 0.001% ponceau S, pH 7.2 supplemented with protease inhibitor cocktail III (Calbiochem).

Blocking agent

5% non-fat dry milk for 1 h.

Primary incubation

1:100 dilution in PBST + 1% milk overnight at 4°C.

Secondary incubation

HRP conjugated goat anti-mouse (Biorad) for 1 h at room temperature.

Detection

ECL and radiographic film.
