This is a knockout-validated antibody summary, based on the publication "AMPK antagonizes hepatic glucagon-stimulated cyclic AMP signalling via phosphorylation-induced activation of cyclic nucleotide phosphodiesterase 4B", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Figure 1. Immunoblotting AMPK level with GAPDH as loading control in primary hepatocytes from wild-type mice or mice bearing a full-body deletion of the AMPK beta 1 subunit (AMPK beta1-/-). Representative immunoblots are shown, from figure S2 [1].
Antibody information

Antibody: AMPK beta1

Catalog number: AF2854

Summary: Goat polyclonal IgG against E. coli-derived recombinant human AMPK beta 1 (Met1-Ile270). Detects human, mouse, and rat AMPK beta 1 in western blot and immunohistochemistry (paraffin). In western blots, no cross-reactivity with recombinant human AMPK beta 2 is observed.

Company: R&D Systems

Validation Method

Western blot

Sample

Primary hepatocytes from wild-type mice or mice bearing a full-body deletion of the AMPK beta 1 subunit (AMPK beta1 -/- ) were treated with 10 uM 991 (or DMSO as vehicle) prior to stimulation with 10 nM glucagon (Glg) for 15 min.

Blocking agent

Tris-buffered saline (TBS) containing 0.1% (v/v) Tween and 5% (w/v) BSA

Primary incubation

1:1,000 dilution, overnight at 4 C

Detection

ECL Classico substrate (Merck Millipore)
