Validation Method

Western blot | Immunohistochemistry

Sample

Control and conditional Aif-KO mice pneumocytes and lung tumors.

Primary incubation

1:1,000 overnight with the primary antibodies at 4 °C (diluted in 2.5% BSA in 1× TBST buffer).

Secondary incubation

WB: 1:5,000 dilution horseradish peroxidase-conjugated secondary Abs (Promega) at room temperature for 1 h.

IHC: 1:500 dilution peroxidase-conjugated anti-rabbit IgG (DAKO, P0448).

Detection

Enhanced chemiluminescence (GE Healthcare, RPN2106).
