ADRM1 antibody | knockout validation | Santa Cruz sc-166754

This is a knockout-validated antibody summary, based on the publication "Early and consistent overexpression of ADRM1 in ovarian high-grade serous carcinoma", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

ADRM1 antibody | knockout validation | Santa Cruz sc-166754 figure 1
Figure 1. ADRM1 knockout HCT116 cell line and its parental line were probed for RPN13 expression by both IHC and Western blot. No staining in knockout cells were seen by either assay. From [1].
Antibody information

Mouse monoclonal IgG2a κ (kappa light chain)

Company: Santa Cruz

Antibody: ADRM1

Catalog number: sc-166754

Summary: Mouse monoclonal IgG2a κ (kappa light chain) against amino acids 1-205 mapping at the N-terminus of human ADRM1. Recommended for detection of ADRM1 of mouse, rat and human origin by western blot, immunoprecipitation, immunofluorescence, immunohistochemistry (paraffin) and ELISA.

Validation Method

Western blot | Immunohistochemistry

Sample

WT and ADRM1 knockout HCT116 cell lines. WB: cells were lysed using 1 mL of M-PER Mammalian Protein Extraction Reagent for ~100 μL of cells and processed as per manufacturer’s instructions (Pierce, ThermoFisher Scientific, Rockford IL). IHC: RPN13 was stained by following PowerVision Poly-HRP IHC Detection system protocol (Leica Biosystem).

Primary incubation

IHC: 1:500 dilution in Antibody Dilution Buffer (ChemMate) at room temperature for 45 min.

Secondary incubation

IHC: PowerVision Poly-HRP Anti-Mouse IgG for 30 min at room temperature.

Detection

IHC: DAB chromogen (3, 3’-diaminobenzidine tetrahydrochloride; Sigma) for 20 min in the dark.
