Sample

WT and CRISPR-mediated ABCB1-knockout KBV200 and HCT-8/V cells. Cells were harvested and lysed in RIPA buffer (1% NP-40, 0.5% sodium deoxycholate, 0.1% SDS, 10 ng/ml PMSF, 0.03% aprotinin, 1 μM sodium orthovanadate) at 4°C for 30 minutes.

Blocking agent

5% BSA.

Secondary incubation

Horseradish peroxidase-conjugated secondary antibodies.

Detection

Chemiluminescent detection reagents and films.
