ABCA4 antibody | knockout validation | Novus Biologicals NBP1-30032

This is a knockout-validated antibody summary, based on the publication "An ABCA4 loss-of-function mutation causes a canine form of Stargardt disease", as cited below [1]. Labome curates formal publications to compile a list of antibodies with unambiguous specificity within Validated Antibody Database (VAD).

Company: Novus Biologicals

Antibody: ABCA4

Catalog number: NBP1-30032

Summary: Mouse monoclonal IgG1 against partially purified bovine 220 kDa disc rim protein. Reacts with human, mouse, bovine, canine, feline and Xenopus. Suitable for western blot, immunocytochemistry/immunofluorescence, immunohistochemistry, immunohistochemistry (frozen).

Validation Method

Immunohistochemistry

Sample

Control and ABCA4-/- dog retina. Tapetal fundus from the affected male (LAB4), his unaffected heterozygous sibling (LAB6), and an unaffected 10-year-old female Labrador retriever (LAB26) were fixed in 4% PFA in 1x PBS on ice for 15 minutes, washed in 1x PBS for 10 minutes on ice, and cryoprotected in 30% sucrose overnight at 4°C.

Blocking agent

1% donkey serum, 0.02% thimerosal, and 0.1% Triton X-100 in 1x PBS) for 30 minutes at room temperature.

Primary incubation

1:500 dilution at 4°C overnight.

Secondary incubation

1:2,000 dilution Alexa 568 secondary antibody (A10037, Invitrogen, Thermo Fisher Scientific) solution for at least 2 hours at room temperature.

Detection

Fluorescence images were captured using a Zeiss Axioplan 2 microscope equipped with an AxioCam HRc camera.
