Spinal cord of Eif4ebp1-/- and wild-type mice (WT) treated with 4EGI-1 (10 ug, i.t. once a day for 3 days) or vehicle. Ice-cold homogenization buffer containing (in mM): 20 Tris-HCl, pH 7.4; 150 NaCl; 1 EDTA; 1% Triton, 5 NaF; 1.5 Na3VO4, and protease inhibitor cocktail (complete, EDTA-free, Roche Applied Science).

Blocking agent

5% dry milk powder in Tris-buffered saline containing 0.1% Tween-20 (TBS-T) for 1 hr

Primary incubation

1:1000 dilution, overnight

Secondary incubation

1 h with HRP-conjugated secondary antibody,

Detection

Enhanced Chemiluminescence reagent (Perkin Elmer), and exposed to an autoradiography film (Denville Scientific Inc.).
