RIAM (a Rap1-GTP-interacting adapter molecule) defines a family of adaptor molecules that contain a RA-like (Ras association) domain, a PH (pleckstrin homology) domain, and various proline-rich motifs (1). RIAM directly binds to Ena/VASP proteins, important modulators of cell migration that localize to focal adhesions and stress fibers, and co-localizes with them at focal adhesions in response to Ras activation. Moreover, RIAM directly binds to activated Ras in a phosphoinositide-dependent manner. RIAM is believed to be the first direct link between Ras signalling and cytoskeletal remodelling via Ena/VASP proteins during cell migration and spreading (2).
