The sequence of the first four N-terminal amino acids was determined and was found to be Ala-Thr-Pro-Val Biological ActivityBiological Activity is evidenced by high affinity binding of mammalian leptins at 1:1 molar ratio.
purity :
Greater than 98.0% as determined by: (a) Analysis by SEC-HPLC. (b) Analysis by SDS-PAGE.
form :
The protein was lyophilized from a concentrated (1mg/ml) solution with 0.0045mM NaHCO3. Sterile Filtered White lyophilized (freeze-dried) powder.
storage stability :
Lyophilized Leptin Binding Domain although stable at room temperature, should be stored desiccated below 0 degree C. Reconstituted Leptin is best stored refrigerated at 4 degree C.Please prevent freeze-thaw cycles.
other info2 :
Solubility: The lyophilized Leptin Binding Domain is very soluble in water and most aqueous buffers below and above the isoelectric point. Protein Content: Protein quantitation was carried out by two independent methods1. UV spectroscopy at 280 nm using the absorbency value of 2.45 as the extinction coefficient for a 0.1% (1mg/ml) solution. This value is calculated by the PC GENE computer analysis program of protein sequences (IntelliGenetics). 2. Analysis by RP-HPLC, using a calibrated solution of Leptin Binding Domain as a Reference Standard.

products description :
Description: Leptin Binding Domain Human Recombinant also called Leptin soluble receptor produced in E Coli is a single, non-glycosylated, polypeptide chain containing 208 amino acids and having a molecular mass of 24.5 kDa. Leptin Receptor consists of the cytokine binding domain of leptin receptor amino acids 428-635 of human leptin receptor. The Leptin Binding Domain is purified by proprietary chromatographic techniques.

ncbi summary :
The protein encoded by this gene belongs to the gp130 family of cytokine receptors that are known to stimulate gene transcription via activation of cytosolic STAT proteins. This protein is a receptor for leptin (an adipocyte-specific hormone that regulates body weight), and is involved in the regulation of fat metabolism, as well as in a novel hematopoietic pathway that is required for normal lymphopoiesis. Mutations in this gene have been associated with obesity and pituitary dysfunction. Alternatively spliced transcript variants encoding different isoforms have been described for this gene. It is noteworthy that this gene and LEPROT gene (GeneID:54741) share the same promoter and the first 2 exons, however, encode distinct proteins (PMID:9207021).[provided by RefSeq, Nov 2010]
uniprot summary :
LEPR: the receptor for leptin (obesity factor). A single-transmembrane-domain receptor of the cytokine receptor family. On ligand binding, mediates signaling through JAK2/STAT3. Involved in the regulation of fat metabolism and, in a hematopoietic pathway, required for normal lymphopoiesis. May play a role in reproduction. Five alternatively spliced transcript variants have been reported. Protein type: Receptor, cytokine; Membrane protein, integral. Chromosomal Location of Human Ortholog: 1p31. Cellular Component: extracellular region; plasma membrane; integral to membrane; receptor complex. Molecular Function: identical protein binding; hematopoietin/interferon-class (D200-domain) cytokine receptor activity; protein binding; transmembrane receptor activity. Biological Process: cholesterol metabolic process; cell surface receptor linked signal transduction; negative regulation of hydrolase activity; multicellular organismal development; negative regulation of gluconeogenesis; cytokine and chemokine mediated signaling pathway; energy reserve metabolic process. Disease: Leptin Receptor Deficiency
