Western Blot Analysis: 0.5-2 ug/ml detects 50 ng of recombinant MAPKAP Kinase 2, active, but not 50 ng of recombinant MAPKAP Kinase 2, inactive. May not effectively immunoblot MAPKAP Kinase 2 from cell lines and tissues (lot dependant). Phosphorylated MAPKAP Kinase 2 was detected using 1 ug/ml of antibody. Recombinant, inactive MAPKAP K2, 50ng, and recombinant, active MAPKAP K2, 50ng, were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-MAPKAP Kinase 2 (Thr222) (0.5 ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Non-radioactive Kinase Assay: 0.02units of p38a/SAPK2a, active phosphorylated 500 ng of MAPKAP Kinase 2, inactive.
