UsageText :
Suitable for use in ELISA, Western Blot, Neutralization and Immunohistochemistry. ELISA: 0.5-1 ug/ml (detection limit is ~6 ng/well). Western Blot: 1-2 ug/ml (detection limit is ~50 ng/lane under both non-reducing and reducing conditions) Because the antibody is a total IgG fraction, complete monospecificity cannot be assumed. Neutralization: ND50:1-3 ug/ml (0.3u/ml rhEPO in cell line TF-1). To measure the ability of the antibody to neutralize the bioactivity of rhEPO on TF-1 cells, rhEPO was incubated with various concentrations of the antibody for 1 hour at 37?C in a 96 well microplate. Following this preincubation period, TF-1 cells were added. The assay mixture, in a total volume of 100 ul/well, containing antibody at the concentrations indicated, rhEPO at a concentration of 0.3units/ml, and cells at 1x10^5cells/ml, was incubated for 72 hours in a 5% CO2 humidified incubator and pulsed with 3H-thymidine for the final four hours. The cells were then harvested onto glass fiber filters and the 3H-thymidine incorporated into DNA was determined. Immunohistochemistry: 2-15 ug/ml for 5-15um thick, paraffin embedded sections.
