Specificity :
On Western blots of lysates derived from total rat or mouse brain, this monoclonal antibody detects a protein of ~30kD corresponding to the authentic Connexin 30 (Cx30) protein. A Cx30 dimer is not detected although other unidentified bands migrating between 70-80kD may be. When used to probe Western blots of rat liver (rat liver does not contain Cx30), several unknown bands are detected at ~110, 53 and 35kD. Immunostaining of fixed, frozen adult rat brain with CX30-8E8 produces a typical punctate staining pattern in the globus pallidus/hypothalamus. Reacts with Connexin 30 from mice and rats.

Immunohistochemistry: LS-B2112 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2112 was determined to be 2.5 ug/ml. Western Blot (mouse and rat): 1 ug/ml; Detects a protein of ~30kD corresponding to the authentic Connexin 30 (Cx30) protein. A Cx30 dimer is not detected although other unidentified bands migrating between 70-80kD may be observed. When used to probe Western Blots of rat liver (rat liver does not contain Cx30), several unknown bands are detected at ~110, 53 and 35kD. Immunofluorescence (frozen sections): 1-5 ug/ml.
