Background :
PKR is an interferon-inducible serine/threonine-specific proteinkinase. It is widely expressed in eukaryotic organisms and activated by double-stranded RNA. The activation of PKR is accompanied by autophosphorylation at multiple sites. Phosphorylation of Thr446 and Thr451 in the PKR activation loop is required in vivo and in vitro for high-level kinase activity. PKR phosphorylates the alpha subunit of eukaryotic initiation factor 2 and inhibits protein synthesis. It is an important component of the interferon response to virus infection. Recently, it has been reported that PKR also phosphorylates human p53 on serine 392.
Immunogen :
Synthetic peptide derived from the region of PKR that contains threonine451. The sequence was aligned to human.
Specificity :
Phosphorylation site-specific antibody selective for the phosphorylated form of the PKR (double-stranded RNA-dependent protein kinase) protein containing a phosphate on threonine 451. This antibody recognizes a protein of 65-68 kDa resolved by SDS-PAGE, identified as PKR.
