Background :

A cell-permeable dihydropyrazinyl-thioacetamide compound that acts a specific, reversible and ATP-competitive inhibitor of MARK (IC 50 = 3.6 uM) with minimal activity against closely related AMPK, Cdc2/B, Cdk2, Cdk5, GSK-3beta, MARKK, p38 and SAD-B kinases. Shown to reduce endogenous MARK/Par-1 activity in NGF-differentiated PC12 cells and in primary cortical neurons from E18 rats, and decrease microtubule-associated proteins hyperphosphorylation (pTau-Ser 262 ) in CHO-ECFP-MARK2 cells and restore microtubule network at 20 uM.
