Background :
DOG1 (Discovered on GIST 1) Gen encodes a cell surface protein of unknown function, which is exprimed especially in gastrointestinal stromal tumours (GIST). The expression in GIST is independent of the expression of c-kit and PDGFR-alpha mutations. For this reason DOG1 antibody can detect a higher percentage of GIST (87%) than traditionally used markers like CD117 (c-kit)(74%) and CD34, which are used for the differential diagnoses of gastrointestinal stromal tumours (Espinosa et al., 2008). DOG1 immunoreactivity was 79% in GIST with PDGFRa mutations, while only 9% of these were CD117-positive. Therefore the diagnosis with DOG1 could also influence the therapy with Imatinib (Glivec®) a tyrosinkinase inhibitor, which is used for the treatment of CML and GIST.
