Flow (Cell Surface): V alpha 24 J alpha 18 TCR Antibody (6B11) [NBP2-00267] - A cell surface stain was performed on CD3+ hPBMCs with V alpha 24 J alpha 18 TCR antibody (6B11) NBP2-00267PE (top image) and a matched isotype control NBP2-27287 (bottom image). Cells were incubated in an antibody dilution of 1 ug/mL for 20 minutes at room temperature. Both antibodies were conjugated to Phycoerythrin. A co-stain was also performed using CD3 antibody NBP2-24867APC.

A specific band was detected for T Cell Receptor alpha Chain-V alpha 24-J alpha Q at approximately 38 kDa (as indicated). This experiment was conducted under reducing conditions and using Immunoblot Buffer Group 8 .

Figure 1. Western blot analysis of TCR alpha using anti-TCR alpha antibody (A05315). Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. Lane 1: human CCRF-CEM whole cell lysates， . Lane 2: rat spleen tissue lysates, . Lane 3: mouse spleen tissue lysates. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TCR alpha antigen affinity purified polyclonal antibody (Catalog # A05315) at 0.5 ug/mL overnight at 4°C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (Catalog # EK1002) with Tanon 5200 system. A specific band was detected for TCR alpha at approximately 39KD. The expected band size for TCR alpha is at 16KD.
