Human

Mucin-1 (MUC1) is a membrane protein present on normal human breast epithelial cells and cell lines derived from breast carcinomas. Human MUC1 is also localised on the surface of the human milk fat globule. MUC1 is a differentiation marker and specific breast epithelial marker in normal and neoplastic mammary development. It is upregulated in the lactating breast and in carcinomas.

Clone also called "3.14.A3" & "F314AE11". Clone HMFG2 requires very specific culture conditions. Under stress the hybridoma switches it's light chain from lambda to kappa : Recover a vial into a T25 flask into RPMI containing 10% serum. Stand the flask upright and after a few hours (about 4-5) if the cells look large, round and healthy lay it on its side adding more medium if necessary. The cells should be monitored every day and split 1:3 before they too dense. The cells should be relatively large, round and shiny. When they are under stress they quickly become smaller and slightly granular – if this happens it is really too late to do anything and another vial should be recovered. When the cells have been moved up to large flask T180 the same regime should be followed but I used to split 1 flask 1:3 and leave the other 2 flask an extra day or two before harvesting the medium.
