MELAN-A was originally identified by functional screening of expression libraries using melanoma-reactive cytolytic T lymphocytes isolated from melanoma patients. Its expression is restricted to melanin-producing cells, including normal and transformed skin melanocytes and retinal pigment epithelium. Approximately 90% of primary and 80% of metastatic melanoma tumours express MELAN-A transcripts.
