Upon activation by Janus kinases, Stat6 translocates to the nucleus where it regulates cytokine-induced gene expression. Stat6 is activated via phosphorylation at Tyr641 and is required for responsiveness to IL-4 and IL-13 (1-4). In addition, Stat6 is activated by IFNa in B cell lines, where it forms transcriptionally active complexes with Stat2 and p48 (5,6). Protein phosphatase 2A is also involved in regulation of IL-4-mediated Stat6 signaling (7). Applications: Suitable for use in Western Blot, Immunoprecipitation, Immunofluorescence and Flow Cytometry. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Immunoprecipitation: 1:100 Immunofluorescence (IF-IC): 1:100 requires methanol permeabilization Flow Cytometry: 1:100 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
