GDP-Dissociation Inhibitors (GDIs) modulate GTPase activity by binding to small G-Proteins, and regulating GDP/GTP exchange. Members of this family of proteins include Rho-GDI (GDI-1), D4-GDI (GDI-2) and Rhoto Rho and Rab, and D4-GDI, which was recently described as -GDI-3 (GDI-3). Rho-GDI, which can bind being a substrate for Caspase 3 (CPP32/YAMA/Apopain) during Fas-induced apoptosis, are both localized in the cytosol. In contrast, GDI-3 may associate with the membrane and cytoskeleton, primarily binding to RhoB and RhoG. Rho-GDI and D4-GDI are highly homologous, differing primarily in the first 25 amino acids of the N-Terminus. Applications: Suitable for use in Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilutions: Western Blot: 0.5-1ug/ml detects Rho-GDI in RIPA lysates from human A431, HL-60, HeLa, Hs294T, Jurkat and murine 3T3/NIH cells. 3T3 cell lysate was resolved by electrophoresis, transferred to nitrocellulose and probed with R198-20 (0.5ug/ml). Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: 4ug immunoprecipitates Rho-GDI from 500ug of Jurkat RIPA lysate. Optimal dilutions to be determined by the researcher. Included Positive Antigen Control: F4209: Fibroblast Cell, 3T3/A31 lysate. Add 2.5ul 2-mercaptoethanol/100ul of lysate. Boil for 5 minutes to reduce the preparation. Load 20ug of reduced lysate per lane for Western Blot analysis. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
