PTEN (phosphatase and tensin homologue deleted on chromosome ten)/MMAC (mutated in multiple advanced cancers) phosphatase is a tumor suppressor gene implicated in a wide variety of human cancers. PTEN encodes the 403 amino acid polypeptide originally described as a dual-specificity protein phosphatase. The main substrates of PTEN are inositol phospholipids generated by the activation of the phosphoinositide 3 kinase (PI3K). PTEN is a major negative regulator of the PI3K/Akt signaling pathway. PTEN possesses a C-terminal noncatalytic regulatory domain that contains three phosphorylation sites (Ser380, Thr382 and Thr383), which regulates its stability and could play an important role in control of its biological activity. Applications: Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications have not been tested. Recommended Dilution: Western Blot: 1:1000 Immunoprecipitation: 1:50 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
