Protein tyrosine residues are phosphorylated as a result of intracellular protein kinase activation (e.g. via growth factors) during normal growth and development, and in oncogenesis. The most abundant population of target proteins for tyrosine phosphorylation are cell surface glycoproteins. Antibody to phosphotyrosine enables the detection, isolation and characterization of proteins containing phosphotyrosine. Also prevents internalization of activated receptors, e.g. EGFr, when microinjected into cells. The affinity of this clone for phosphotyrosine is approximately 10e-6 to 10e-7 M. Binding of this Ab to phosphorylated tyrosines can be inhibited by free phosphotyrosine and phenylphosphate, but not by phosphoserine, phosphothreonine or free phosphate.1-3 Applications: • Detection of phosporylated tyrosine-containing proteins on Western blots 1 • Immunoaffinity purification of tyrosine-phosporylated proteins 1 • Quantitation of phosphotyrosine by Enzyme-Linked-Immunosorbent-Assay (ELISA)1 • Immunofluorescence analysis 4 Recommended Dilution: Western Blot: 2ug/ml Immunoaffinity Purification: 4mg/ml ELISA: 100ng/ml Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Store at -20°C or colder. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
