Desc1 :

Phosphorylation and dephosphorylation of cellular proteins are central steps in transducing extracellular signals to the cell nucleus. Phosphorylated epitopes may serve as docking sites for the assembly of protein complexes or may alter the 3-dimensional protein structure thus modulating enzymatic activity or the ability to undergo protein-protein-interactions. Modification of proteins on serine residues is mediated by serine/threonine kinases. Applications: Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested. Recommended Dilutions: ELISA: 0.05ug/ml Western Blot: 1ug/ml for HRPO/ECL detection. Recommended blocking buffer: BSA/Tween20 based buffer. Western Blot (BPPT): Recommended blocking buffer: 3% BSA, 1% PEG 4000, 1% PVP, 0.1% Tween20, 2x PBS. Do not use milk of casein for blocking. Immunoprecipitation: 1-10ug per 10e6 pervanadate-treated A431 cells. Optimal dilutions to be determined by the researcher. Positive Control Cell Lysate (Included): P4076-22A: Phosphoserine Positive Control. Use 20ul/lane. Storage and Stability: May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 12 months. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
