Phosphorylation of tyrosine residues is associated with many growth factor receptors and oncogene protein kinases, and is an important signal for cellular activation, proliferation and differentiation. Antibodies to phosphotyrosine have proven to be useful in the characterization of these kinase substrates that may be involved in oncogenesis and regulation of cell growth.
Applications:
Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications not tested.
Recommended Dilution:
Western Blot: 1:1000
Immunoprecipitation: 2-10ug/IP reaction
Note: Binding is inhibited by high salt concentrations and by divalent cations. Use a buffer with less than 150mM sodium chloride and divalent cations not much greater than 1mM.
Optimal dilutions to be determined by the researcher.
Positive Control:
Mouse brain, NIH 3T3 cells (+/-TPA), K562 cells, EGF-stimulated A431 cells and MCF-7 cell lysates.
Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
