Specific reversible protein phosphorylation plays a central role in regulating many cellular events by controlling the activity of participating proteins. Ways that phosphorylation/dephosphorylation can modulate a protein’s activity include altering its conformation, localization, stability, and ability to associate or form complexes with other proteins(3–5). As much as one third of cellular proteins may be phosphorylated at any given moment. Reversible protein tyrosine phosphorylations are central to regulating events in signal transduction pathways initiated by most growth factors and cytokines.
Applications:
Suitable for use in Immunoprecipitation. Other applications not tested.
Recommended Dilution:
Immunoprecipitation: 25–50 ul/IP
This antibody can be used for immunoprecipitation and column affinity chromatography. One ml of agarose can bind to about 1 mg of tyrosine phosphorylated proteins found in normal tissues and cells. Before use, rinse agarose to eliminate 1 mM phosphotyrosine. This antibody’s binding is inhibited by high salt concentrations and by divalent cations. Use a buffer with less than 150 mM NaCl and divalent cations of not much greater than 1 mM.
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage and to avoid repeated freezing and thawing, aliquot and add glycerol (40-50%). Freeze at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.

This antibody will react specifically to tyrosine phosphorylated proteins in both the native and denatured states. Binding is inhibited by phosphotyrosine and will not react with serine or threonine phosphorylated residues on proteins.
