Phosphatidylcholine-specific phospholipase D (PLD) hydrolyzes phosphatidylcholine (PC) to produce choline and phosphatidic acid (PA). PA is the precursor of the second messenger diacylglycerol (DAG). Two isoforms of PLD (PLD1 and PLD2) have been identified. Both are regulated by protein kinases, small GTPases and Ca2+ (1). PLD1 is phosphorylated at serine 2, serine 561 and threonine 147 by PKC (2,3). Phosphorylation at threonine 147 and serine 561 regulates PLD1 activity (3). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
