Phosphoinositide-dependent protein kinase 1 (PDK1) plays a central role in many signal transduction pathways, activating Akt, PKC isoenzymes p70 S6 kinase and RSK. Through its effects on these kinases, PDK1 is involved in regulation of a wide variety of processes, including cell proliferation, differentiation and apoptosis. Several serine sites (Ser25, Ser241, Ser393/396 and Ser410) are phosphorylated on PDK1 in unstimulated human embryo kidney 293 cells, as well as IGF-1 stimulated cells (4). Phosphorylation on the activation loop Ser241 by autophosphorylation is necessary for PDK1 activity. Applications: Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications have not been tested. Recommended Dilution: Western Blot: 1:1000. Incubate membrane with diluted antibody in TBS, 5% BSA, 0.1% Tween-20 at 4°C with gentle shaking, overnight. Immunoprecipitation: 1:100 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
