N-methyl-D-aspartate receptor (NMDAR) mediates long-term potentiation and slow postsynaptic excitation, which play central roles in learning, neurodevelopment and neuroplasticity (1). The receptor function can be regulated by the protein kinase/phosphatase complex associated with it (2). PKC can phosphorylate the NR1 subunit (NMDAR1) of the receptor on Ser890/ Ser896 and PKA can phosphorylate NR1 on serine 897 (3). The phosphorylation of NR1 by PKC decreases its affinity to calmodulin binding, thus preventing the inactivation effect of calmodulin on NMDAR (4). The phosphorylation of NR1 by PKA probably counteracts the inhibitory effect of calcinurin on the receptor (5). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
