The mammalian target of rapamycin, mTOR, also know as FRAP or RAFT (1-3), is a serine/ threonine protein kinase. mTOR acts as a sensor for ATP, amino acids, balancing the availability of nutrients and cell growth (4,5). When sufficient nutrients are available, mediated by phosphatidic acid (6), mTOR transmits a positive signal to p70 S6 kinase and participates in the inactivation of the eIF4E inhibitor 4E-BP1. These events result in activation of the translation of specific mRNA subpopulations. mTOR is phosphorylated at serine 2,448 via the PI3 kinase/Akt signaling pathway and autophosphorylated at serine 2,481 (6,7).
Applications:
Suitable for use in ELISA, Western Blot and Immunoprecipitation. Other applications have not been tested.
Recommended Dilution:
Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight.
Immunoprecipitation: 1:100
Optimal dilutions to be determined by the researcher.
Storage and Stability:
May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
