Mammalian Ste20-like kinases, including Mst1-4, belong to the germinal center kinase (GCK) family. The N-terminal kinase domain of Mst is considerably homologous to the kinase domain of yeast Ste20 kinase and other p21-activated kinases (1). The C-terminal region of Mst1 and Mst2 contains a dimerization and an inhibitory domain, while the C-terminal region of Mst3 and Mst4 are not well characterized (1-3). Dimerization and phosphorylation at the activation loop regulates translocation of Mst1 from the cytosol to the nucleus (3). Growing evidence indicates that Mst1 and Mst2 are activated by apoptotic signals as well as other stress conditions (4-6). The full activation of Mst1 requires both phosphorylation and caspase-mediated cleavage (4). Activated Mst kinases may rely on p38 MAPK and JNK pathways to amplify apoptotic signals (5). Inhibitory phosphorylation is also reported. Phosphorylation at serine 327 of Mst1, which is close to the caspase-3 recognization site, inhibits caspasemediated cleavage (4). Applications: Suitable for use in Western Blot. Other applications not tested. Recommended Dilution: Western Blot: 1:1000 Optimal dilutions to be determined by the researcher. Storage and Stability: May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 12 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
